DETERMINATION OF FUNCTIONAL SPECIFICATIONS AND BIOACTIVE COMPONENTS FOR SOME ORIGANUM SPECIES

The genus Origanum is represented in Turkey by 24 species, the ratio of endemism in the genus is 63 %. In this research, essential oils of Origanum vulgare, Origanum onites and Origanum minutiflorum oregano species which are obtained from different sources, have been isolated using the hydrodistillation process and components has been analysed by a GC-MS system. In this study the antioxidant and the antimicrobial properties of essential oil samples were also examined.
The gas chromatography-mass spectrometry analyses revealed that the major components of essential oils for Origanum minutiflorum are carvacrol (% 67.86 -84.95), p-cymen (% 5.36 8.77) and -terpinen (% 0.99 3.46). The major components of essential oil detected from Origanum onites are carvacrol (% 30.01 -71.96), thymol (% 1.53 38.25), p-simen (% 3.90 10.07) and -terpinen (% 2.25 5.07). And examining the essential oil composition of the Origanum vulgare (993) samples harvested in Alanya, it has identified that % 81.35 carvacrol, % 10.17 linalool, % 1.27 p-cymen, % 1.21 thymol and % 1.01 -caryophyllen are the main components.
In this study, the antioxidant activities were investigated by using DPPH (1,1 diphenyl2-picryl-hydrazyl) free radical scavenging activity. EC50 values of the extracts of Origanum species in the study were identified using DPPH method. It was found that these values varied between 0,44 16,21 L and O. onites had the lowest value (0,44 L) while O. vulgare had the highest value (16,21 L).
Also, essentail oils were plated at different concentrations (5, 10, 20, 30, 40, 50, and 80 L/mL) by mixing bacterial cultures and the number of live bacteria were determined . The results revealed that Origanum essential oils exhibited a very strong antibacterial effect on Escherichia coli O157:H7 and reduced the counts of Listeria monocytogenes indeed (P<0.05).
